Application Notes
Suitable for use in Flow Cytometry, ELISA and Western Blot.
Dilution: Intracellular Flow Cytometry: 25ug/ml; Use 10ul to label 1-5x10e5 cells in a total reaction volume not exceeding 200ul. Cells must be fixed and permeablized using 4% paraformaldehyde and 0.1% saponin in PBS.
Direct ELISA: 0.5-1.0ug/ml; The detection limit for recombinant human mGluR5 is ~2ng/well
Western Blot: 1-2ug/ml; The detection limit for recombiant human mGluR5 is ~25ng/lane under non-reducing and reducing conditions.

Recombinant protein corresponding to aa19-509 from human mGluR5, expressed in NSO cells (P41594).

Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute by adding sterile PBS. Aliquot and store at -20 degree C. Reconstituted product is stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Human metabotropic glutamate receptor 5 (mGluR5; also mGluR5b) is a 150kD, 7-transmembrane glycoprotein that belongs to group I of the C-family of G-protein coupled receptors. mGluR5 is constitutively expressed and regulates neuronal ion channel activity. Human mGluR5 is 1212aa in length and contains an extracellular domain (ECD) of 558aa. Through its ECD, mGluR5 either homodimerizes or heterodimerizes with the Ca++-sensor receptor. There is one alternate splice form (mGluR5a) that shows a 32aa deletion between aa877-908 in the cytoplasmic tail. Over aa19-509, human mGluR5 is 98% identical to mouse, rat and canine mGluR5.
