Specificity
Specific for p27 when phosphorylated at Thr187. On Western blots, it identifies the target band at ~27kD. It does not react with unphosphorylated p27. Specificity has been confirmed with Thr187-mutant p27 recombinant protein. Reactivity has been confirmed with p27-transfected 293 cells. Based on sequence homology, this antibody may also react with mouse, rat and hamster.

Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

The cyclin-dependent kinase inhibitor p27KIP1 is a potent inhibitor of both Cyclin E-Cdk2 and Cyclin A-Cdk2, two cdk complexes involved in the regulation of G1/S transition. p27 is phosphorylated on threonine 187 by cyclin E/Cdk2, and overexpression of this complex in mammalian cells induces degradation of transfected p27. The Cdk-mediated phosphorylation of p27 on threonine 187 is required for p27 ubiquitination and consequent degradation. The phosphorylation on threonine 187 and the ubiquitination of cellular p27, as well as p27 ubiquitinating activity, were found to be regulated collectively during the cell cycle, being present in proliferating cells but not in G0- and G1- arrested cells.

This gene encodes a cyclin-dependent kinase inhibitor, which shares a limited similarity with CDK inhibitor CDKN1A/p21. The encoded protein binds to and prevents the activation of cyclin E-CDK2 or cyclin D-CDK4 complexes, and thus controls the cell cycle progression at G1. The degradation of this protein, which is triggered by its CDK dependent phosphorylation and subsequent ubiquitination by SCF complexes, is required for the cellular transition from quiescence to the proliferative state. Mutations in this gene are associated with multiple endocrine neoplasia type IV (MEN4). [provided by RefSeq, Apr 2014]
