p21Cip1/WAF1 (p21) is an inhibitor of cyclin-dependent kinases (CDK) and is transcriptionally activated by p53. The p21 promoter contains a p53-binding site and transcriptional activation can occur through wild-type but not mutant p53. Induction of p21 has been shown to be essential for p53-mediated arrest of the cell cycle in G1 in response to DNA damage, and transfection of p21 into both tumor cell lines and normal fibroblasts results in growth inhibition. Although expression of p21 has been demonstrated to be largely dependent upon p53, p53-independent expression of p21 has also been observed. Simultaneously with its discovery as a p53 regulated gene, p21 was also shown to be a potent inhibitor of G1 cyclin dependent kinases. P21 can associate with every cyclin kinase complex examined; however, the protein appears to preferentially inhibit CDK/cyclin complexes that play a role in the G1/S transition. Thus, p21 has been postulated to act predominantly as a cell cycle buffer which must be titrated in order to exit the G1 phase of the cell cycle. In addition to its role as a CDK inhibitor, p21 has been shown to bind and inactivate components of the DNA replication machinery.
