Western Blot (WB)
(Western blot analysis: Protein samples (MDA-MB-435 cell lysates, conditioned medium from MDA-MB-435 cells containing secreted OPN or recombinant protein) were resolved by SDS-PAGE on a 8-16% gradient gel, transferred to nitrocellulose, and blocked for 2 hrs in Tris-buffered saline containing 0.05% Tween 20 (TBS-T) and 5% milk powder. For immunodetection, Blots were incubated with anti-OPN 53 (5µg/ml) overnight at 4 degree C, followed by incubation with a HRP-conjugated goat anti-mouse secondary antibody (in TBS-T) for 1 hr at room temperature. Protein bands were visualized by enhanced chemiluminescence on Kodak X-OMAT film. M= Magic Mark.)

The protein encoded by this gene is involved in the attachment of osteoclasts to the mineralized bone matrix. The encoded protein is secreted and binds hydroxyapatite with high affinity. The osteoclast vitronectin receptor is found in the cell membrane and may be involved in the binding to this protein. This protein is also a cytokine that upregulates expression of interferon-gamma and interleukin-12. Several transcript variants encoding different isoforms have been found for this gene.
