Western Blot (WB) (Western Blot: Detection of Human Olig1 by Western Blot. Western blot shows lysates of Neuro­2A mouse neuroblastoma cell line and SH­SY5Y human neuroblastoma cell line. PVDF membrane was probed with 0.5ug/ml of Human Olig1 Monoclonal Antibody followed by HRP­conjugated Anti­Mouse IgG Secondary Antibody. A specific band was detected for Olig1 at ~28kD (as indicated). Application Notes
Suitable for use in ELISA, Western Blot, and Immunohistochemistry.
Dilution: Western Blot: 0.5ug/ml
Immunohistochemistry: 8-25ug/ml detects Olig1 in paraffin- embedded human astrocytoma and glioma tissue sections.
ELISA: 0.5-1ug/ml. The detection limit for recombinant human Olig1 is ~0.5ng/ml.
Immunogen
Recombinant cooresponding to human Oligodendrocyte transcription factor-1, expressed in E. coli (XM_170977).
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Immunohistochemistry (IHC)

(Immunohistochemistry Olig1 in Human Glioma. Olig1 was detected in immersion fixed paraffin­ embedded sections of human glioma using 8ug/ml Human Olig1 Monoclonal Antibody overnight at 4°C. Tissue was stained with the Anti­ Mouse HRP­DAB Cell & Tissue Staining Kit (brown) and counterstained with hematoxylin (blue).)

Olig1 and Olig2, which have been implicated in oligodendrogenesis, are expressed in the region of the ventral ventricular zone of late embryonic spinal cord where oligodendrocyte progenitors appear. Olig3 is transiently expressed in different types of progenitors of embryonic central nervous system and then disappears in the course of development. In adult, expression of Olig3 is primarily detected in neural tissues.
