ELISA: 0.5-1ug/ml; Detection limit for rhNotch-2 ICD is ~12ng/well.

Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol, PBS, and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Human Notch-2 is a 300kD member of the notch family of transmembrane (TM) proteins. It is synthesized as a 2446 amino acid (aa) type I TM glycoprotein that undergoes Golgi processing to generate a 180kD disulfide-linked extracellular domain (ECD) (likely aa25-1538) and a 110kD membrane bound truncated segment (likely aa1539-2471). Upon binding to Delta1, Jagged1, or Jagged2, the 110kD segment undergoes two cleavages, the second which generates an NICD (notch intracellular domain) (aa1697-2471), a potential nuclear transcription factor. In the nucleus, NICD regulates CSL pathways. When NICD is phosphorylated, it is deactivated.

NCBI Description

This gene encodes a member of the Notch family. Members of this Type 1 transmembrane protein family share structural characteristics including an extracellular domain consisting of multiple epidermal growth factor-like (EGF) repeats, and an intracellular domain consisting of multiple, different domain types. Notch family members play a role in a variety of developmental processes by controlling cell fate decisions. The Notch signaling network is an evolutionarily conserved intercellular signaling pathway which regulates interactions between physically adjacent cells. In Drosophilia, notch interaction with its cell-bound ligands (delta, serrate) establishes an intercellular signaling pathway that plays a key role in development. Homologues of the notch-ligands have also been identified in human, but precise interactions between these ligands and the human notch homologues remain to be determined. This protein is cleaved in the trans-Golgi network, and presented on the cell surface as a heterodimer. This protein functions as a receptor for membrane bound ligands, and may play a role in vascular, renal and hepatic development.
