Suitable for use in Western Blotting, Immunoprecipitation
Immunoprecipitation: 4ug immunoprecipitats Nibrin/Nbs1 from 500ug of Jurkat RIPA lysate. Jurkat cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti- Nibrin/Nbs1 (2ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Nibrin/Nbs1 (95kD).
Immunogen
GST fusion protein corresponding to residues 420-774 of human Nibrin.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Immunofluorescence (IF)

(Immunofluorescent analysis of A431 cells was performed using 2ug/mL of MBS601230 and visualized with a Donkey anti-Mouse IgG secondary antibody conjugated to Cy3 (Left and Right image) (Red). Actin filaments have been labeled with Alexa Fluor® 488-conjugated phalloidin (Left and Right image) (Green). Nucleus is stained with DAPI (Right image) (Blue). This antibody positively stains the nucleus.)

Mutations in this gene are associated with Nijmegen breakage syndrome, an autosomal recessive chromosomal instability syndrome characterized by microcephaly, growth retardation, immunodeficiency, and cancer predisposition. The encoded protein is a member of the MRE11/RAD50 double-strand break repair complex which consists of 5 proteins. This gene product is thought to be involved in DNA double-strand break repair and DNA damage-induced checkpoint activation. [provided by RefSeq]
