Application Notes
Suitable for use in ELISA, Western Blot and Immunoprecipitation.
Dilution: Direct ELISA: 0.5-1ug/ml. Detection limit for rhChymase is ~1ng/well.
Western Blot: 1-2ug/ml. Detection limit for rhChymase is
~10ng/lane under non-reducing and reducing conditions. Chemiluminescent detection wil increase sensitivity by 5 to 50 fold.
Immunoprecipitation: 25ug/ml from conditioned cell culture medium.

Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Chymase-1 (also mast cell protease I) is a secreted, 30kD glycoprotein member of the granzyme subfamily, peptidase S1 family of enzymes. It is secreted by mast cells and vascular smooth muscle cells, and converts angiotensin I into angiotensin II. This promotes fibroblast proliferation and secretion of ECM. Mature human chymase is 226aa in length. It contains one peptidase S1 domain (aa 2-245). There is one potential alternate splice form. It shows a deletion of the 99 C-terminal amino acids (aa149-247).

NCBI Description

This gene product is a chymotryptic serine proteinase that belongs to the peptidase family S1. It is expressed in mast cells and thought to function in the degradation of the extracellular matrix, the regulation of submucosal gland secretion, and the generation of vasoactive peptides. In the heart and blood vessels, this protein, rather than angiotensin converting enzyme, is largely responsible for converting angiotensin I to the vasoactive peptide angiotensin II. Angiotensin II has been implicated in blood pressure control and in the pathogenesis of hypertension, cardiac hypertrophy, and heart failure. Thus, this gene product is a target for cardiovascular disease therapies. This gene maps to 14q11.2 in a cluster of genes encoding other proteases. [provided by RefSeq]
