Purified bovine 300 kDa CI-MPR. Epitope: This antibody is shown to recognize an epitope in the extracellular domain of MPR.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot Store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Lysosomal enzymes containing one or two mannose 6-phosphate (man6P) moieties are moved about in the cell by two distinct but interconnected cycles by means of 300kD cation independent mannose 6-phospate receptors (CI-MPR). The biosynthetic cycle refers to the MPR transport of newly synthesized lysosomal enzymes from the trans Golgi network to late endosomes or early lysosomes. The endocytic cycle transports extracellular lysosomal enzymes from the plasma membrane via clathrin-coated vesicles to early endosomes. The entire pool of MPRs cycles between these cellular compartments every 3 hours. The steady state distribution of MPR's is predominantly within late endosomes, fewer in the trans Golgi network and ~10 % at cell surface.

NCBI Description

The protein encoded by this gene is a receptor for both, insulin-like growth factor 2 (IGF2) and mannose 6-phosphate (M6P). The IGF2 and M6P binding sites are located on different segments of the receptor. This receptor functions in the intracellular trafficking of lysosomal enzymes, the activation of transforming growth factor beta, and the degradation of IGF2. While the mouse Igf2r gene shows exclusive expression from the maternal allele, imprinting of the human IGF2R gene appears to be polymorphic, with only a minority of individuals showing expression from the maternal allele. IGF2R: a multifunctional type I transmembrane protein receptor that binds insulin-like growth factor 2 at the cell surface and mannose-6-phosphate-tagged proteins in the trans-Golgi network. Also known as the cation-independent mannose-6 phosphate (M6P) receptor. Clears IGF2 from the cell surface to attenuate signaling, and transports M6P-tagged lysosomal enzymes from the Golgi complex and the cell surface to lysosomes. M6P-tagged lysosomal enzymes bind specifically to mannose-6- phosphate receptors in the Golgi apparatus and the resulting receptor-ligand complex is transported to an acidic prelyosomal compartment where the low pH mediates the dissociation of the complex. Acts as a positive regulator of T-cell coactivation, by binding DPP4. Belongs to the MRL1/IGF2R family.
