Immunoprecipitation (IP)

(MD1L1 was immunoprecipitated from 1mg of Jurkat cells membrane fraction, blotted with M25102 of 10 ug. Western blot was performed from the immunopre-cipitate using M25102 at 1/1000 dilution. Anti-Mouse-IgG(HRP), specific to the non-reduced form of IgG, was used as secondary antibody at 1/10000 dilution. Lane 1: Jurkat cells membrane fraction. Lane2: IP product of Jurkat cells membrane fraction.Predicted band size : 83 KDaObserved band size : 83 KDaBlocking and dilution buffer and concentration:5% milk/TBST.)

Flow Cytometry (FC/FACS)

(Flow cytometric analysis of 2% paraformaldehyde-fixed Jurkat (Human T cell leukemia cells from peripheral blood) cells labeling MD1L1 with M25102 at 1/200 dilution (red) compared with a mouse monoclonal IgG isotype control (black) and an unlabelled control (cells without incubation with primary antibody, blue). Goat anti-mouse IgG (FITC) at 1/300 dilution was used as the secondary antibody.)

MAD1L1 functions as a homodimer and interacts with MAD2L1. MAD1L1 may play a role in cell cycle control and tumor suppression. Alternative splicing results in multiple transcript variants. Uniprot Description

Component of the spindle-assembly checkpoint that prevents the onset of anaphase until all chromosomes are properly aligned at the metaphase plate. May recruit MAD2L1 to unattached kinetochores. Has a role in the correct positioning of the septum. Required for anchoring MAD2L1 to the nuclear periphery. Binds to the TERT promoter and represses telomerase expression, possibly by interfering with MYC binding.
