Application Notes
Application Use: For Western blot and immunohistology dilutions to be used depend on detection system applied. It is recommended that users test the reagent and determine their own optimal dilutions. The typical starting working dilution is 1:50. Fixation with PFA or acetone/methanol is recommended.

Product should be stored at 4 degree C. Under recommended storage conditions, product is stable for one year.

Product Description: Afadin is an actin filament-binding protein that is involved in cell-cell adherens junctions (AJs) which directly interacts with the alpha herpus virus receptor, a member of the immunoglobulin supergene family. Afadin is colocalized with nectin at cadherin-based cell-cell adherence junctions in various tissues and cell lines. L-Afadin has been isolated as an F-actin-binding protein from rat brain. L-afadin is ubiquitously expressed and specifically localized at zonula adherens (ZA) in epithelial cells and at cell-cell adherens junctions (AJs) in non-epithelial cells. L-afadin has a splice variant, named small intestine absorptive afadin (s-afadin) which is abundantly expressed in neural tissue. Monoclonal antibody 3 has been raised against peptide 1814-1829 aa of rat l-afadin. Monoclonal antibody 3 cross reacts with human, mouse and dog l-afadin.

Afadin: plays a role in adhesion by participating, along with E-cadherin and catenin, in the organization of homotypic, interneuronal and heterotypic cell-cell adherens junctions (AJs). Essential for the association of nectin and E-cadherin. Contains 1 forkhead-associated (FHA) domain, a phosphopeptide recognition domain; 1 dilute (DIL) domain, a myosin-like domain which may play a functional role in cellular processes or vectorial vesicle transport; 1 PDZ domain, which frequently binds the carboxyl-terminal sequences of proteins or phosphatidylinositol 4,5-bisphosphate in the plasma membrane; and 2 RA domains, which are associated with RasGTP effector activity. Five alternatively spliced isoforms have been described. Isoform 2 does not interact with F-actin.
