Applicable Applications for anti-KERA antibody
Western Blot (WB), Immunohistochemistry (IHC), ELISA (EIA)
Application Notes
ELISA; Western Blot: recommended dilution 0.4 mg/mL; IHC
Note
This epitope is best recognized after keratanase and keratanase II treatment/diges-tion of section (IHC) or tissue extracts (for Western analyses). In some IHC tissue analyses, chondroitinase treatment may also be used as this second enzyme treatment facilitates antibody penetration into the tissue.
Preparation and Storage
Store at -20°C. Avoid freeze/thaw cycles.

Related Product Information for anti-KERA antibody
Keratocan belongs to the Small Leucine Rich Proteoglycan (SLRP) family. It was originally found to be a cornea specific keratan sulphate proteoglycan that, along with Lumican, plays a role in the development and maintenance of corneal transparency. Mutations of keratocan cause cornea plana in humans, which is often associated with glaucoma, whereas the absence of Lumican leads to formation of cloudy corneas. This suggests that Keratocan and Lumican have different roles in regulating the formation of stromal extracellular matrix.

NCBI Description

The protein encoded by this gene is a keratan sulfate proteoglycan that is involved in corneal transparency. Defects in this gene are a cause of autosomal recessive cornea plana 2 (CNA2).[provided by RefSeq, May 2010]

Uniprot Description

Function: May be important in developing and maintaining corneal transparency and for the structure of the stromal matrix.

Subcellular location: Secreted › extracellular space › extracellular matrix

By similarity.

Tissue specificity: Cornea. Increased expression in the stroma of keratoconus corneas. Also detected in trachea, and in low levels, in intestine, skeletal muscle, ovary, lung and putamen. Ref.4

Post-translational modification: Binds keratan sulfate chains

By similarity.

Involvement in disease: The autosomal recessive cornea plana 2 (CNA2) [MIM:217300]: In CNA2, the forward convex curvature is flattened, leading to a decrease in refraction, reduced visual activity, extreme hyperopia (usually plus 10 d or more), hazy corneal limbus, opacities in the corneal parenchyma, and marked arcus senilis (often detected at an early age). CNA2 is a rare disorder with a worldwide distribution, but a high prevalence in the Finnish population.Note: The disease is caused by mutations affecting the gene represented in this entry. Ref.2 Ref.6

Sequence similarities: Belongs to the small leucine-rich proteoglycan (SLRP) family. SLRP class II subfamily.Contains 10 LRR (leucine-rich) repeats.Contains 1 LRRNT domain.

Product Notes

The KERA kera (Catalog #AAA442013) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's Keratocan can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC), ELISA (EIA). ELISA; Western Blot: recommended dilution 0.4 mg/mL; IHC. Researchers should empirically determine the suitability of the KERA kera for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Keratocan, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
