• Application Notes: Research Application(s): ELISA Detection: MP4-25D2 monoclonal antibody is useful as a detection antibody in a sandwich ELISA for quantifying human IL-4 protein levels.1,3 Biotinylated MP4-25D2 antibody should be paired with a suitable purified antibody as the capture antibody, with purified recombinant human IL-4 as the standard.; ; Immunofluorescence/Flow Cytometry: MP4-25D2 antibodies are useful for intracytoplasmic staining and flow cytometric analysis to identify and enumerate IL-4-positive cells within mixed cell populations. Recombinant human IL-4, unlabeled MP4-25D2, or Biotinylated Rat IgG1 should be used as a specificity control.; ; Western Blotting: The purified MP4-25D2 antibody is also useful for Western blotting applications. A concentration of 1-5 ug/mL (in conjunction with AP-labeled Goat Anti-Rat-Ig) has been found to enable detection of <= 100 ng/lane of recombinant human IL-4 under reducing conditions.; ; Neutralization: The LE/AF form of MP4-25D2 can be used for neutralizing human IL-4 bioactivity. A suitable LE/AF Rat IgG1 should be used as an isotype control.; ; Since applications vary, each investigator should determine the optimal concentration appropriate for individual applications.
• Immunogen: CHO-expressed recombinant human IL-4
• Characterization: To ensure acceptable performance, each batch of product is tested in a sandwich ELISA to conform to characteristics of a standard reference reagent.
• Preparation and Storage: The purified (UNLB) antibody is supplied as 0.5 mg purified immunoglobulin in 1.0 mL of 100 mM borate buffered saline, pH 8.2. No preservatives or amine-containing buffer salts added. Store at 2- 8 degree C. ; The biotin (BIOT) conjugate is supplied as 0.5 mg labeled antibody in 1.0 mL of PBS/NaN3. Store at 2-8 degree C. ; The low endotoxin, azide-free (LE/AF) antibody is supplied as 0.5 mg purified immunoglobulin in 1.0 mL of PBS. Aliquot and store at or below -20 degree C. ; Protect conjugated forms from light. Aliquot and freeze the low endotoxin, azide-free product at -20 degree C immediately upon receipt. Each reagent is stable for the period shown on the bottle label if stored as directed.

Uniprot Description

Function: Participates in at least several B-cell activation processes as well as of other cell types. It is a costimulator of DNA-synthesis. It induces the expression of class II MHC molecules on resting B-cells. It enhances both secretion and cell surface expression of IgE and IgG1. It also regulates the expression of the low affinity Fc receptor for IgE (CD23) on both lymphocytes and monocytes.
