Application Notes
ELISA: In a sandwich ELISA (assuming 100ul/well), a concentration of 3-4 ug/ml of this antibody will detect at least 0.1 ng/well of recombinant murine IL-12 when used with PeproTech's biotinylated antigen affinity purified anti-murine IL-12 as the detection antibody at a concentration of at least 0.5ug/ml.
Antibody Generation
Monoclonal antibodies were produced in rats using recombinant murine IL-12 as the immunizing antigen. The IgG antibody was purified from ascites fluid followed by Protein G affinity chromatography.
Neutralization
To yield one-half maximal inhibition [ND50] of the biological activity of mIL-12 (2.0 ng/ml), a concentration of 0.16 - 0.24 ug/ml of this antibody is required.
Related Product Information for anti-IL-12 antibody
Interleukin 12 (IL-12), also known as natural killer cell stimulatory factor (NKSF) or cytotoxic lymphocyte maturation factor (CLMF), is a heterodimeric pleiotropic cytokine made up of a 40 kDa (p40) subunit and a 35 kDa (p35) subunit. The IL-12 p40 subunit is shared by IL-23, another heterodimeric cytokine that has biological activities similar to, as well as distinct from, IL-12. IL-12 is produced by macrophages and B cells and has been shown to have multiple effects on T cells and natural killer (NK) cells. While mouse IL-12 is active on both human and mouse cells, human IL-12 is not active on mouse cells.

Product Notes

The IL-12 il12a (Catalog #AAA690325) is an Antibody produced from Rat and is intended for research purposes only. The product is available for immediate purchase. The Rat Anti-Mouse IL-12 reacts with Mouse and may cross-react with other species as described in the data sheet. Researchers should empirically determine the suitability of the IL-12 il12a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IL-12, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
