This gene encodes a soluble cytokine that is a member of the type II interferon class. The encoded protein is secreted by cells of the both the innate and adaptive immune systems. The active protein is a homodimer that binds to the interferon gamma receptor which triggers a cellular response to viral and microbial inflections. Mice deficient in this gene have increased susceptibility to viral, bacterial and parasitic infections and to several autoimmune diseases. [provided by RefSeq, Sep 2015]

Function: Produced by lymphocytes activated by specific antigens or mitogens. IFN-gamma, in addition to having antiviral activity, has important immunoregulatory functions. It is a potent activator of macrophages, it has antiproliferative effects on transformed cells and it can potentiate the antiviral and antitumor effects of the type I interferons.

The Rat Anti-Mouse IFN-g-FITC reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's IFN-g can be used in a range of immunoassay formats including, but not limited to, ELISA. ELISA: XMG1.2 monoclonal antibody can be used as a capture antibody in a sandwich ELISA for quantifying mouse IFN-gamma protein levels. Biotinylated XMG1.2 antibody can be paired with a suitable purified capture antibody, with purified recombinant human IFN-gamma as the standard.; Western Blot: The purified XMG1.2 antibody is also useful for Western Blot applications. For Western Blot, a concentration of 1-5 ug/mL (in conjunction with AP-labeled Goat Anti-Rat-Ig, has been found to enable detection of <= 100 ng/lane of recombinant mouse IFN-gamma under reducing conditions.; Immunofluorescence/Flow Cytometry: XMG1.2 antibody is useful for intracytoplasmic staining and flow cytometric analysis to identify and enumerate IFN-gamma-positive cells within mixed cell populations.4-6 Recombinant mouse IFN-gamma or Rat IgG1 should be used as a specificity control. FITC conjugated XMG1.2 antibodies are useful for intracellular staining in cells producing IFN-gamma. Intracellular staining buffers are recommended for this application.; Neutralization: The LE/AF form of XMG1.2 can be used for neutralizing mouse IFN-gamma bioactivity. A suitable LE/AF Rat IgG1 should be used as an isotype control.; Since applications vary, each investigator should determine the optimal concentration appropriate for individual applications. Researchers should empirically determine the suitability of the Ifng ifng for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IFN-g, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening.
