Western Blot (WB) (Western Blot analysis of HLA Class 1 ABC on Hela lysate. Proteins were transferred to a nitrocellulose membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody was used at a 1:500 dilution in 5% BSA at room temperature for 2 hours, washed in TBST, and probed with an HRP-conjugated secondary antibody for 1 hour at room temperature. Chemiluminescent detection was performed using SuperSignal ECL Western Blotting Substrate.)

Related Product Information for anti-HLA antibody
HLA-B belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta-2 microglobulin). The heavy chain is anchored in the membrane. Class I molecules play a central role in the immune system by presenting peptides derived from the endoplasmic reticulum lumen. They are expressed in nearly all cells. The heavy chain is approximately 45 kDa and its gene contains 8 exons. Exon 1 encodes the leader peptide, exon 2 and 3 encode the alpha1 and alpha2 domains, which both bind the peptide, exon 4 encodes the alpha3 domain, exon 5 encodes the transmembrane region and exons 6 and 7 encode the cytoplasmic tail. Polymorphisms within exon 2 and exon 3 are responsible for the peptide binding specificity of each class one molecule. Typing for these polymorphisms is routinely done for bone marrow and kidney transplantation. Hundreds of HLA-B alleles have been described.

Product Notes

The HLA hla-b (Catalog #AAA3014477) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The HLA Class 1 ABC Monoclonal Antibody reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's HLA Class 1 ABC can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunocytochemistry (ICC). WB: 1:500-1:2000 ICC: 1:50-1:200. Researchers should empirically determine the suitability of the HLA hla-b for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "HLA Class 1 ABC, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
