Purified, NS0-derived, recombinant human HGF R (rhHGF R; Accession# P08581; aa 25-932) extracellular domain.
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute by adding sterile 40-50% glycerol, aliquot and store at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Human hepatocyte growth factor receptor, (HGF R), a product of the proto-oncogene c-MET, is a heterodimeric transmembrane glycoprotein that is a receptor-type tyrosine kinase. HGF R is synthesized as a 1390 aa single-chain precursor which undergoes cotranslational glycosylation and proteolytic cleavage to give rise to the heterodimeric mature form. The 190kD HGF R heterodimer is composed of a 50kD chain (aa 25-307) disulfide-linked to a 145kD chain (aa 308-1390). The chain is exposed to the cell surface and the chain spans the plasma membrane.

NCBI Description

The proto-oncogene MET product is the hepatocyte growth factor receptor and encodes tyrosine-kinase activity. The primary single chain precursor protein is post-translationally cleaved to produce the alpha and beta subunits, which are disulfide linked to form the mature receptor. Various mutations in the MET gene are associated with papillary renal carcinoma. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq]

Uniprot Description

Met: a proto-oncogenic receptor tyrosine kinase with high affinity for hepatocyte growth factor. The primary single chain precursor protein is post-translationally cleaved to produce the 45 kDa alpha- and 145 kDa beta-subunits, which are disulfide linked to form the mature receptor. Ligand-binding induces autophosphorylation at multiple tyrosines, which recruit several downstream signaling components, including Gab1, c-Cbl and PI3 kinase. Activating point mutations cause hereditary papillary renal carcinoma. Mutations also seen in sporadic renal cell carcinoma and childhood hepatocellular carcinoma. Upregulation in carcinomas and sarcomas correlates with metastasis and poor outcome. Some gastric carcinomas harbor a translocation that creates an activated TPR-Met fusion protein. A small molecule inhibitor (PHA-665752) shows an effect in gastric carcinoma xenografts. Inhibitors: SU11274, PHA-665752, mAbs. Two alternatively spliced human isoforms have been reported.
