Specificity
VU1D9 reacts with the first EGF repeat in the extracellular domain of Ep-CAM. The epitope is sensitive for reducing agents. VU1D9 detects Ep-CAM in most simple, pseudostRatified and transitional epithelia and in the majority of Human carcinomas.

Each vial contains 100 ul 1 mg/ml purified monoclonal antibody in PBS containing 0.09% sodium azide.

Application Notes
VU1D9 is suitable for immunoblotting (non-reducing conditions), immunocytochemistry and immuno-histochemistry on frozen and paraffin-embedded tissues and flow cytometry. Optimal antibody dilution should be determined by titRation; recommended range is 1:25 - 1:200 for flow cytometry, and for immunohistochemistry with avidin-biotinylated Horseradish peroxidase complex (ABC) as detection reagent, and 1:100 - 1:1000 for immunoblotting appliCations.

VU1D9 is a Mouse monoclonal IgG1 antibody derived by fusion of Mouse myeloma cells with spleen cells from a Mouse immunized with an NCI-H69 small cell lung carcinoma cell line.

Ep-CAM is a 40 kD glycoprotein and can be detected at the basolateral membrane of the majority of epithelial tissues, where it is intriCately linked with the cadherin-Catenin complex and hence the fundamental WNT pathway responsible for intracellular signalling and polarity. This antigen functions as a homotypic calcium-independent cell adhesion molecule. Of particular interest, Ep-CAM appears to be overexpressed by the majority of Human epithelial carcinomas, including colorectal, breast, prostate, head and neck, and hepatic carcinomas. The antigen is being used as a target for immunotherapy of Human carcinomas. Formation of Ep-CAM-mediated adhesions has a negative regulatory effect on adhesions mediated by classic cadherins, which may have strong effects on the differentiation and growth of epithelial cells. Ep-CAM overexpression was suggested to be associated with enhanced epithelial prolifeRation.

Researchers should empirically determine the suitability of the EPCAM epcam for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Ep-CAM (CD326), Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage.
