Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50?, aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Elk-1, a 62kD transcription factor, is the most abundant component of ternary complex factors that is regulated by phosphorylation and in turn regulates gene expression in response to multiple stimuli. Elk-1 is a substrate of MAP kinase and phosphorylated by ERK2 on several sites in the carboxyl terminus, including serine 383. Phosphorylation of Elk-1 on serine 383 is critical for its transcriptional activity, including Elk-1-mediated transcription of c-Fos. Dephosphorylation of serine 383 by calcineurin downregulates Elk-1 transcriptional activity.

NCBI Description

This gene is a member of the Ets family of transcription factors and of the ternary complex factor (TCF) subfamily. Proteins of the TCF subfamily form a ternary complex by binding to the the serum response factor and the serum reponse element in the promoter of the c-fos proto-oncogene. The protein encoded by this gene is a nuclear target for the ras-raf-MAPK signaling cascade. Alternatively spliced transcript variants encoding the same protein have been found for this gene. ELK1: a transcription factor of the Ets family and the ternary complex factor (TCF) subfamily. Forms a ternary complex by binding to the serum response factor and the serum response element in the promoter of the c-fos proto-oncogene. Mediates gene activity in response to serum and growth factors. Phosphorylated by MAP kinase pathways at a cluster of S/T motifs at its C-terminus; phosphorylation at these sites is critical for transcriptional activation by Elk-1. Two alternatively-spliced isoforms have been described.
