For Western Blots, incubate membrane with diluted antibody in 5% w/v nonfat dry milk, 1X TBS, 0.05% tween-20 at 4°C with gentle shaking overnight.

Immunoprecipitation (IP)

(DBT was immunoprecipitated from 1mg of PC9 cells membrane fraction, blotted with M25355 of 10 ug. Western blot was performed from the immunoprecipitate using M25355 at 1/1000 dilution. Anti-Mouse-IgG(HRP), specific to the non-reduced form of IgG, was used as secondary antibody at 1/10000 dilution. Lane 1: PC9 cells membrane fraction. Lane2: IP product of PC9 cells membrane fraction.Blocking and dilution buffer and concentration:5% milk/TBST.)

The branched-chain alpha-keto acid dehydrogenase complex (BCKD) is an inner-mitochondrial enzyme complex involved in the breakdown of the branched-chain amino acids isoleucine, leucine, and valine.
