Specificity
34betaE12 is specific for "high molecular weight" cytokeratins 1, 5, 10 and 14, corresponding to molecular weights 68, 58, 56.5 and 50 kDa, respectively, which are characteristically found in "complex" epithelium. The antibody reacts with all squamous and ductal epithelium as well as carcinomas such as squamous cell carcinoma and adenocarcinoma of ductal origin (e.g., breast, pancreas) but does not react with sarcoma, melanoma or lymphoma. This antibody reacts with benign small-acinar lesions of the prostate and has been shown to be useful as an adjunct in the identification of prostate carcinoma.

Application Notes
Immunohistochemistry (paraffin sections; for optimal results, fixation in alcohol-based fixatives, such as methacarn, is recommended. Pretreatment with proteolytic enzymes such as trypsin may be required when formalin-fixed sections are to be stained. For optimal staining of prostate tissue higher concentrations of protease and longer incubation times are recommended, e.g., 0.5% trypsin at 37 degree C for 2 to 4 hours. Choose a good quality enzyme-linked anti-mouse IgG1 antibody for detection)

NCBI Description

The protein encoded by this gene is a member of the keratin gene family. The type II cytokeratins consist of basic or neutral proteins which are arranged in pairs of heterotypic keratin chains coexpressed during differentiation of simple and stratified epithelial tissues. This type II cytokeratin is specifically expressed in the spinous and granular layers of the epidermis with family member KRT10 and mutations in these genes have been associated with bullous congenital ichthyosiform erythroderma. The type II cytokeratins are clustered in a region of chromosome 12q12-q13. [provided by RefSeq, Jul 2008]

K1: a type II cytoskeletal keratin. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. There are two types of cytoskeletal and microfibrillar keratin: type I (acidic; 40-55 kDa) [K9 to K20] and type II (neutral to basic; 56-70 kDa) [K1 to K8]. Both a basic and an acidic keratin are required for filament assembly. Generally associates with K10.
