Mouse anti-Human Clusterin Monoclonal Antibody | anti-CLU antibody
Anti-Clusterin Antibody, Mouse Monoclonal
No cross-reactivity in ELISA with Mouse CLU/Clusterin/Apolipoprotein J/Apo-J, Human cell lysate (293 cell line)
ELISA: 1:5000-1:10,000
ICC/IF: 1:100-1:500
FC/FACS/FCM: 1:100-1:500
Immunofluorescence (IF)
(Immunofluorescence staining of Human CLU in Hela cells. Cells were fixed with 4% PFA, permeabilzed with 1% Triton X-100 in PBS, blocked with 10% serum, and incubated with Mouse anti-Human CLU monoclonal antibody (1:300) at 37 degree C 1 hour. Then cells were stained with the Alexa Fluor 488-conjugated Goat Anti-mouse IgG secondary antibody (green). Positive staining was localized to cytoplasm.)
Flow Cytometry (FC/FACS)
(Flow cytometric analysis of Human CLU expression on A549 cells. The cells were treated according to manufacturer's manual (BD Pharmingen'), stained with purified anti-Human CLU, then a FITC-conjugated second step antibody. The fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact cells.)
Western Blot (WB)
(Anti-CLU mouse monoclonal antibody at 1:500 dilutionLane A: Hela Whole Cell LysateLysates/proteins at 30 ug per lane.SecondaryGoat Anti-Mouse IgG H&L (Dylight800) at 1/15000 dilution.Developed using the Odyssey technique. NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is a secreted chaperone that can under some stress conditions also be found in the cell cytosol. It has been suggested to be involved in several basic biological events such as cell death, tumor progression, and neurodegenerative disorders. Alternate splicing results in both coding and non-coding variants.[provided by RefSeq, May 2011]
Uniprot Description
CLU: Isoform 1 functions as extracellular chaperone that prevents aggregation of nonnative proteins. Prevents stress- induced aggregation of blood plasma proteins. Inhibits formation of amyloid fibrils by APP, APOC2, B2M, CALCA, CSN3, SNCA and aggregation-prone LYZ variants (in vitro). Does not require ATP. Maintains partially unfolded proteins in a state appropriate for subsequent refolding by other chaperones, such as HSPA8/HSC70. Does not refold proteins by itself. Binding to cell surface receptors triggers internalization of the chaperone-client complex and subsequent lysosomal or proteasomal degradation. Secreted isoform 1 protects cells against apoptosis and against cytolysis by complement. Intracellular isoforms interact with ubiquitin and SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complexes and promote the ubiquitination and subsequent proteasomal degradation of target proteins. Promotes proteasomal degradation of COMMD1 and IKBKB. Modulates NF-kappa-B transcriptional activity. Nuclear isoforms promote apoptosis. Mitochondrial isoforms suppress BAX-dependent release of cytochrome c into the cytoplasm and inhibit apoptosis. Plays a role in the regulation of cell proliferation. Belongs to the clusterin family. 5 isoforms of the human protein are produced by alternative splicing.

Product Notes
The CLU clu (Catalog #AAA8105243) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Anti-Clusterin Antibody, Mouse Monoclonal reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Clusterin can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), ELISA (EIA), Flow Cytometry (FC/FACS/FCM), Immunocytochemistry (ICC), Immunofluorescence (IF). WB: 1:500-1:1000 ELISA: 1:5000-1:10,000 ICC/IF: 1:100-1:500 FC/FACS/FCM: 1:100-1:500. Researchers should empirically determine the suitability of the CLU clu for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Clusterin, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
