Application Notes
LK2H10 is useful for immunohistochemistry on paraffin embedded and frozen tissues. Optimal antibody dilution should be determined by titRation; recommended dilution 1:10 when applied in the Indirect immunofluorescence method. With the ABC method the dilution can be 5-10x higher. Dilute immediately before use with PBS or TRIS-HCl pH 7,0.

LK2H10 is a Mouse monoclonal IgG1 antibody derived by fusion of NS-1 Mouse myeloma cells with spleen cells from a BABL/c Mouse immunized with an immunogen that has been isolated from Human pheochromocytoma.
Preparation and Storage
Store undiluted antibody at 2-8 degree C (until expiry date). For prolonged storage: store aliquots at -20 degree C.

Neurons and neuroendocrine cells contain membrane-delimited pools of peptide hormones, biogenic amines, and neurotransmitters with a characteristic electron-dense appearance. These vesicles, which are present throughout the neuroendocrine system and in a variety of neurons, store and release chromogranins and secretogranins, a unique group of acidic, soluble secretory proteins. Chromogranin A, a protein of 439 amino acids which is encoded on chromosome 14, and an apparent molecular weight of 68kDa, is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, phenochromocytomas, paragangliomas, and other neuroendocrine tumors, including small cell lung carcinoma.

The protein encoded by this gene is a member of the chromogranin/secretogranin family of neuroendocrine secretory proteins. It is found in secretory vesicles of neurons and endocrine cells. This gene product is a precursor to three biologically active peptides; vasostatin, pancreastatin, and parastatin. These peptides act as autocrine or paracrine negative modulators of the neuroendocrine system. Other peptides, including chromostatin, beta-granin, WE-14 and GE-25, are also derived from the full-length protein. However, biological activities for these molecules have not been shown. CHGA: a member of the chromogranin/secretogranin family of neuroendocrine secretory proteins. It is found in secretory vesicles of neurons and endocrine cells and is a precursor to multiple biologically active peptides including vasostatin, pancreastatin, and chromostatin. Binds calcium with a low-affinity. Vasostatin negatively regulates angiogenesis and has antibacterial activity against some Gram-positive bacteria. Pancreastatin inhibits glucose induced insulin release from the pancreas. Chromostatin inhibits catecholamine release from chromaffin cells.
