Specificity
Recognizes human CHIT1 in direct ELISAs and Western Blots. In these formats, this antibody shows no crossreactivity with rhCHI-3L1. Species sequence homology
Purity/Purification
Affinity Purified
Purified by Protein G affinity chromatography.
Form/Format
Supplied as a lyophilized powder from PBS, 5% trehalose. Reconstitute with 200ul sterile 40-50% glycerol, PBS.
Applicable Applications for anti-Chitotriosidase antibody
ELISA (EL/EIA), Immunoprecipitation (IP), Neutralization
Application Notes
Dilution: ELISA (direct): 0.5-1.0ug/ml. Detection limit for rhCHIT1 is approximately 2ng/well.
Immunoprecipitation: 25ug/ml to immunoprecipitate rhCHIT1 from conditioned cell culture medium.
Neutralization: The antibody was preincubated with the enzyme at different molar ratios at RT for 30 min. The enzyme was then assayed with 4-MU-beta-D-N,N',N''-triacetylchitotriose. 50% of the enzymatic activity was inhibited by the antibody at approximately 56ng/mL (IC 50) under conditions in which the enzyme was present at 20ng/mL and the substrate concentration was 0.01mM. IC 50 was achieved at approximately 1:1 molar ratio of the antibody to the enzyme.
Immunogen
NS0-derived rhCHIT-1
Hybridoma
Fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, NS0-derived, recombinant human CHIT1.
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute by adding sterile 40-50% glycerol, PBS. Aliquot and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Related Product Information for anti-Chitotriosidase antibody
Human CHIT-1 (chitotriosidase-1; also chitinase-1) is a 50kD, secreted glycoprotein that belongs to glycosylhydrolase family #18. It is synthesized by macrophages, and may serve to degrade chitin (poly-N-acetylglucosamine)-containing pathogens such as fungi and nematodes. Mature CHIT-1 is 435 amino acids (aa) in length, and contains a catalytic domain (aa #22-372) followed by a chitin-binding region (aa #373-466). Very high levels of CHIT1 are found in the plasma of patients with Gaucher's disease type I. Mature human CHIT-1 is 79% aa identical to mouse CHIT-1.

Product Notes

The Chitotriosidase (Catalog #AAA603316) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Chitotriosidase (Chitinase 1, CHIT-1) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Chitotriosidase can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Immunoprecipitation (IP), Neutralization. Researchers should empirically determine the suitability of the Chitotriosidase for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Chitotriosidase, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
