Mouse CD81 (Mr 25 kDa), a member of the transmembrane 4 integral membrane protein family
CD81/TAPA-1 is an integral membrane protein expressed on a variety of cell types, and has a high degree of sequence homology between human and mouse.1 CD81 is expressed on thymic stromal cells where it plays an important role in the transition of gamma delta + T cells to more mature T cells with alpha beta T cell receptors.2 Immunohistochemical staining has revealed that its expression is localized to the subcapsular region of the thymus and, specifically, on cells that have distinct clustering patterns. It has been speculated that the ligand for CD81 is the pre-T cell receptor, which is composed of a TCR beta chain and glycoprotein pT alpha.2 The monoclonal antibody 2F7 can block thymocyte interaction with CD81 in vitro.2

Characterization
To ensure lot-to-lot consistency, each batch of monoclonal antibody is tested by flow cytometry to conform to characteristics of a standard reference reagent. Representative data are included in this product insert.
Preparation and Storage
The purified (UNLB) antibody is supplied as 0.5 mg of purified immunoglobulin in 1.0 mL of 100 mM borate buffered saline, pH 8.2. No preservatives or amine-containing buffer salts added. Store at 2- 8 degree C.
The fluorescein (FITC) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8 degree C.
The biotin (BIOT) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8 degree C.
The R-phycoerythrin (R-PE) conjugate is supplied as 0.1 mg in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C. Do not freeze!
Protect conjugated forms from light. Each reagent is stable for the period shown on the bottle label if stored as directed.
The low endotoxin, azide-free (LE/AF) antibody is supplied as 0.5 mg purified immunoglobulin in 1.0 mL of PBS. Aliquot and store at or below -20 degree C.
Protect conjugated forms from light. Aliquot and freeze the low endotoxin, azide-free product at -20 degree C immediately upon receipt. Each reagent is stable for the period shown on the bottle label if stored as directed.

NCBI Description

The protein encoded by this gene is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Most of these members are cell-surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the regulation of cell development, activation, growth and motility. This encoded protein is a cell surface glycoprotein that is known to complex with integrins. This protein appears to promote muscle cell fusion and support myotube maintenance. Also it may be involved in signal transduction. This gene is localized in the tumor-suppressor gene region and thus it is a candidate gene for malignancies. [provided by RefSeq]

Function: May play an important role in the regulation of lymphoma cell growth. Interacts with a 16-kDa Leu-13 protein to form a complex possibly involved in signal transduction. May acts a the viral receptor for HCV.

Subunit structure: Plays a critical role in HCV attachment and/or cell entry by interacting with HCV E1/E2 glycoproteins heterodimer. Interacts directly with IGSF8. Ref.7 Ref.8 Ref.9

Subcellular location: Membrane; Multi-pass membrane protein.

Tissue specificity: Hematolymphoid, neuroectodermal and mesenchymal tumor cell lines.

Involvement in disease: Defects in CD81 are the cause of immunodeficiency common variable type 6 (CVID6) [

MIM:613496]; also called antibody deficiency due to CD81 defect. CVID6 is a primary immunodeficiency characterized by antibody deficiency, hypogammaglobulinemia, recurrent bacterial infections and an inability to mount an antibody response to antigen. The defect results from a failure of B-cell differentiation and impaired secretion of immunoglobulins; the numbers of circulating B cells is usually in the normal range, but can be low. Ref.10
