Specificity
The antibody RIV11 is directed against human CD8 and recognises all CD8 positive T-cells, thymocytes and natural killer cells.

Each vial contains 500 ul 0.2 mg/ml purified monoclonal antibody in phosphated buffered saline (PBS) containing 50% glycerol + 0.2% bovine serum albumin (BSA) + 0.02% sodium azide.

Application Notes
The RIV11 antibody is suitable for the detection of total CD8 by flow cytometry and ELISA.

The antibody is also suitable for the detection of CD8 by immunocytochemistry and immunohistochemistry on frozen tissues. Human lymph node, spleen and peripheral blood lymphocytes are suitable for use as positive controls.

Optimal antibody dilutions for the different applications should be determined by titration.

RIV11 is a mouse monoclonal IgG1 antibody derived by fusion of 653 mouse myeloma cells with splenocytes from a BALB/c mouse immunized with human peripheral blood lymphocytes.

CD8 is a 32kDa cell surface transmembrane glycoprotein expressed by most thymocytes, a subpopulation of mature peripheral blood T-cells and natural killer (NK) cells. CD8 is expressed on the cell surface as a heterodimer, comprising two distinct disulphide linked alpha and beta polypeptide chains, or as a CD8alpha homodimer.

The CD8 alphabeta heterodimer is expressed on T-cell receptor (TCR) alphabeta+ cytotoxic T-lymphocytes (CTL's), whereas CD8alpha homodimers are expressed on TCR gammadelta+ T-cells, natural killer cells and a subpopulation of TCR alphabeta+ T-cells. The CD8 molecule functions as a co-receptor (with the T-cell receptor) as part of the antigen-recognition complex on TCRalphabeta+, class I MHC-restricted T cells and plays an important role in T-cell signaling. The CD8 Ig-like domain binds to the alpha-3 domain of MHC-Class 1.

NCBI Description

The CD8 antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell-cell interactions within the immune system. The CD8 antigen acts as a coreceptor with the T-cell receptor on the T lymphocyte to recognize antigens displayed by an antigen presenting cell in the context of class I MHC molecules. The coreceptor functions as either a homodimer composed of two alpha chains or as a heterodimer composed of one alpha and one beta chain. Both alpha and beta chains share significant homology to immunoglobulin variable light chains. This gene encodes the CD8 alpha chain. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Nov 2011]

CD8A: Identifies cytotoxic/suppressor T-cells that interact with MHC class I bearing targets. CD8 is thought to play a role in the process of T-cell mediated killing. CD8 alpha chains binds to class I MHC molecules alpha-3 domains. In general heterodimer of an alpha and a beta chain linked by two disulfide bonds. Can also form homodimers. Shown to be expressed as heterodimer on thymocytes and as homodimer on peripheral blood T-lymphocytes. Interacts with the MHC class I HLA-A/B2M dimer. Interacts with LCK in a zinc-dependent manner. 2 isoforms of the human protein are produced by alternative splicing.
