• Application Notes: 1:50 (or use 1.6ul of concentrated stock per 5 x 105-cell test)
• Performance: Five x 10⁵ cultured Raji human tumor cells were washed and incubated 45 minutes on ice with 80 ul of anti-CD70/R-PE at a 1:50 dilution (10 ug/ml). Cells were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.33 log₁₀ fluorescent units when compared to a Mouse IgG1/PE negative control (MBS666704) at a similar concentration. Binding was blocked when pre incubated 10 minutes with 20 ul of 0.5 mg/ml anti-CD70 antibody (MBS666352)
• Production: Protein A purified antibody from tissue culture supernatant was conjugated to R-Phycoerythrin through a sulfo-ester linkage. Unconjugated antibody aPE were removed using size exclusion chromatography. Antibody conjugate is at 0.5 mg/ml with PE: Ab molar ratio of 1.21.
• Immunogen: Human WM-1 (Waldenström’s macroglobulinemia) cell line

Information: Human CD70 is not present on resting lymphocytes. CD70 is expressed after activation on about 20% of T cells and on about 70% of B cells. CD70 is a ligand for CD27 and it is also a member of the tumor necrosis factor gene family. Antibody BU69 recognizes CD70 molecule of approximately 30 kd. Antibody BU69 inhibits T cell proliferation induced by dendritic cells.

The protein encoded by this gene is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. This cytokine is a ligand for TNFRSF27/CD27. It is a surface antigen on activated, but not on resting, T and B lymphocytes. It induces proliferation of costimulated T cells, enhances the generation of cytolytic T cells, and contributes to T cell activation. This cytokine is also reported to play a role in regulating B-cell activation, cytotoxic function of natural killer cells, and immunoglobulin sythesis. [provided by RefSeq, Jul 2008]
