Performance
Five x 105 cultured HEL92.1 cells were washed and incubated 45 minutes on ice with 80 mul of anti-CD69/Biotin at 10 mug/ml. Cells were washed twice and incubated with 2 degree reagent Streptavidin/R-Phycoerythrin, after which they were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.06 log10 fluorescent units when compared to a Mouse IgG2a/Biotin negative control at a similar concentration. Binding was blocked when cells were pre incubated 10 minutes with 20 mul of 0.5 mg/ml anti-CD69 antibody.

Antibody from (low FBS containing) tissue culture supernatant was Protein A purified to >95% mouse immunoglobulin by SDS-PAGE (<1% bovine immunoglobulin), and reacted with NHS-Biotin. Unconjugated Biotin was removed from conjugate using a desalting column.

50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl, 5% Glycerol, 0.2% BSA, 0.04% NaN3 (as a preservative).

Information: Human CD69 is expressed by platelets and mature thymocytes and appears rapidly after activation on T cells, B cells and NK cells. This pattern of cellular CD69 expression has suggested a lymphocyte activation role. Functional studies with antibodies to CD69 also support an activation role for CD69. Antibody HP-4B3 recognizes the dimeric CD69 molecule of 60 kd.
