Mouse Anti-Human CD33-UNLB
CD33 is a 67 kDa type I transmembrane glycoprotein and a member of the sialoadhesin family of cell surface receptors. It is absent from pluripotent stem cells but appears on myelomonocytic precursors after CD34. It then continues to be expressed on both the myeloid and monocyte lineages, although it is absent on granulocytes. While it has been reported that CD33 can function as a sialic acid-dependent cell adhesion molecule, cells expressing CD33 require desialylation before they can bind cells bearing the appropriate sialoglycoconjugates. This suggests that inhibitory cis interactions may regulate or block any adhesion function.1-4
The fluorescein (FITC) conjugate is supplied as 100 tests in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C.
The R-phycoerythrin (R-PE) and allophycocyanin (APC) conjugates are supplied as 100 tests in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C. Do not freeze!
The Spectral Red (SPRD) and R-phycoerythrin-Cyanine 5.5 (R-PE-CY 5.5) conjugates are supplied as 100 tests in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C. Do not freeze!
The low endotoxin, azide-free (LE/AF) antibody is supplied as 0.5 mg purified immunoglobulin in 1.0 mL of PBS. Aliquot and store at or below -20 degree C.
With the exception of the low endotoxin, azide-free product, the vials should be stored at 2-8 degree C. Conjugated forms should not be frozen and should be protected from prolonged exposure to light. Aliquot and freeze the low endotoxin, azide-free product at -20 degree C immediately upon receipt. Each reagent is stable for the period shown on the bottle label if stored as directed.

Function: Putative adhesion molecule of myelomonocytic-derived cells that mediates sialic-acid dependent binding to cells. Preferentially binds to alpha-2,6-linked sialic acid. The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface. In the immune response, may act as an inhibitory receptor upon ligand induced tyrosine phosphorylation by recruiting cytoplasmic phosphatase(s) via their SH2 domain(s) that block signal transduction through dephosphorylation of signaling molecules. Induces apoptosis in acute myeloid leukemia (in vitro). Ref.5 Ref.7

Post-translational modification: Phosphorylation of Tyr-340 is involved in binding to PTPN6 and PTPN11. Phosphorylation of Tyr-358 is involved in binding to PTPN6.
