Notes
The following is a general protocol, optimization is required for best results of your experiment.)
1. Take 100 ul peripheral blood anticoagulated by EDTA and add to the bottom of 5 ml tube
2. Add <=1.0ug labeled antibody to the bottom of flow tube mixing with the whole blood, incubate for 20 minutes at room temperature away from light
3. Add 2 ml 1×RBC lysis buffer, incubate for 10 minutes away from light after mixing, dissolve red blood cells
4. Sample tube is set to 1000 rpm centrifugation for 5 minutes, discard the supernatant
5. Add 2 ml PBS wash buffer to resuspend the cells, then 1000 rpm centrifugation for 5 minutes, discard the supernatant
6. Add 0.5 ml PBS wash buffer to resuspend the cells and detect by flow cytometry (sample should be tested on the day on the machine, or be added fixation overnight at 4 degree C then measured); [PBS wash buffer: PBS +1% FBS +0.1% NaN3] [Cell fixation: 2% formaldehyde solution]
Preparation and Storage
Store at 4 degree C. DO NOT freeze. Light sensitive.
Related Product Information for anti-CD3 antibody
CD3 is a complex composed of delta, gamma, epsilon, and zeta chains. They are 20-25 kD members of the immunoglobulin superfamily and associated with the T cell receptor (TCR).CD3 is expressed on thymocytes, peripheral T cells, some NK-T cells, and dendritic epidermal T cells. CD3 is involved in antigen recognition, signal transduction, and T cell activation.

Product Notes

The CD3 cd3e (Catalog #AAA4753698) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The CD3 mouse mAb (PE) reacts with Rat and may cross-react with other species as described in the data sheet. AAA Biotech's CD3 can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS). Researchers should empirically determine the suitability of the CD3 cd3e for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CD3, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
