CD19 is a human protein encoded by the CD19 gene. CD19 is expressed on follicular dendritic cells and B-cells; it is present on B-cells from earliest recognizable B-lineage cells during development to B-cell blasts, but is lost on maturation to plasma cells. In normal lymphoid tissue, CD19 is observed in germinal centers (on both B-cells and follicular dendritic cells), in mantle-zone cells, and in scattered cells in the interfollicular areas, with an overall immunoreactivity pattern similar to that ofCD20 and CD22. However, in contrast to CD20, CD19 is also expressed in pre-B-cells. CD19 positivity is seen in the vast majority of B-cell neoplasms (B-Lymphoblastic Lymphoma, Small Lym-phocytic Lymphoma/CLL, Mantle Cell Lymphoma, Follicular Lymphoma, Burkitt's Lymphoma, Marginal Zone Lymphoma, Diffuse Large B-cell Lymphoma, T-cell-rich B-cell Lymphoma, Lymphoblastic Lymphoma, Hairy Cell Leukemia), and commonly at a lower intensity than normal B-cell elements. Plasma cell neoplasms are consistently negative, as are T-cell neoplasms. CD19 expression is not seen in Reed-Sternberg cells of classic Hodgkin's Disease.

NCBI Description

Lymphocytes proliferate and differentiate in response to various concentrations of different antigens. The ability of the B cell to respond in a specific, yet sensitive manner to the various antigens is achieved with the use of low-affinity antigen receptors. This gene encodes a cell surface molecule which assembles with the antigen receptor of B lymphocytes in order to decrease the threshold for antigen receptor-dependent stimulation. Uniprot Description

Function: Assembles with the antigen receptor of B lymphocytes in order to decrease the threshold for antigen receptor-dependent stimulation.

Subunit structure: Forms a complex with CD21, CD81 and CD225 in the membrane of mature B cells. Interacts with VAV. Interacts with GRB2 and SOS when phosphorylated on Tyr-348 and/or Tyr-378. Interacts with PLCG2 when phosphorylated on Tyr-409. Ref.11

Subcellular location: Membrane; Single-pass type I membrane protein.

Post-translational modification: Phosphorylated on serine and threonine upon DNA damage, probably by ATM or ATR. Phosphorylated on tyrosine following B-cell activation. Ref.10 Ref.11 Ref.12

Involvement in disease: Defects in CD19 are the cause of immunodeficiency common variable type 3 (CVID3) [

MIM:613493]; also called antibody deficiency due to CD19 defect. CVID3 is a primary immunodeficiency characterized by antibody deficiency, hypogammaglobulinemia, recurrent bacterial infections and an inability to mount an antibody response to antigen. The defect results from a failure of B-cell differentiation and impaired secretion of immunoglobulins; the numbers of circulating B cells is usually in the normal range, but can be low. Ref.13

Sequence similarities: Contains 2 Ig-like C2-type (immunoglobulin-like) domains.

Sequence caution: The sequence AAA35533.1 differs from that shown. Reason: Frameshift at position 396.
