Performance
Five x 105 ficoll prepared human PBMC per tube were pre incubated with 20 ul of 250 ug/ml human Ig (to block non specific binding) after which they were incubated 45 minutes on ice with 80 ul of product at a dilution factor of 1:50 (10 ug/ml). Cells were washed three times, fixed and analyzed by FACS using a lymphocyte gate. Cells stained positive with a mean shift of 0.67 log10 fluorescent units when compared to a Mouse IgG1/R-PE negative control. Binding was blocked when the reagent was pre incubated with a 10-fold excess of recombinant CD127-muIg.

Protein A purified antibody from tissue culture supernatant was conjugated to R-Phycoerythrin through a sulfo-ester linkage. Unconjugated antibody was removed using size exclusion chromatography. Antibody conjugate is at 0.5 mg/ml with an A565/A280 ratio of 2.64.

Information: CD127 (IL-7Ra) is the specific receptor component for the cytokine Interleukin -7 (IL-7). It is found on a wide variety of hematopoietic cell types including B cell precursors, and the majority of T cells. Its expression levels are decreased on T cells following activation. CD127 can dimerize with CD132(IL-2Rg) to form a high affinity IL-7 receptor (1). CD127 engagement is necessary for T cell development in humans (2). Antibody ANC8F2 recognizes an extracellular epitope of recombinant and native CD127 in FACS and Enzyme Immunoassay.
