Performance
Five x 105 ficoll prepared human peripheral white blood cells were washed and preincubated ~ 10 minutes with 20 ul human IgG at 250 ug/ml (to block non specific binding) after which they were incubated 45 minutes on ice with 80 ul of anti-CD11b/Biotin at a concentration of 10 ug/ml. Cells were then washed twice and incubated 45 minutes with 2 degree reagent Streptavidin/R-PE, after which they were washed three times, fixed and analyzed using a BD FACstar plus. A net 19% sub population of the cells stained positive with a mean shift of 1.69 log10 fluorescent units when compared to a Mouse IgG1/Biotin negative control at a similar concentration. Binding was 94% blocked when cells were preincubated ~10 minutes with 20 ul of 0.5 mg/ml unlabeled anti-CD11b antibody.

Antibody from (low FBS containing) tissue culture supernatant was Protein A purified to >95% mouse immunoglobulin by SDS-PAGE (<1% bovine immunoglobulin), and reacted with NHS-Biotin. Unconjugated Biotin was removed from conjugate using a desalting column.

Information: Human CD11b (alpha M integrin) complexes with CD18 (beta 2 integrin) to form the complement receptor type 3 (CR3) heterodimer which binds to ICAM-1, fibrinogen, C3b, and coagulation factor X. CD11b expression is restricted to leukocytes, mainly to myeloid and NK cells. Antibody ICRF44 recognizes the integrin alpha M subunit (CD11b) of about 165 kd. Antibody ICRF44 blocks homotypic neutrophil and monocyte (FMLP induced) aggregation.
