Application Notes
ICC: 5-10ug/ml of a previous lot showed positive immunostaining for Cbl in 3T3 cells fixed with ice cold methanol/ethanol [1:1] for 1 minute.
IP: 2ug of a previous lot immunoprecipitated Cbl from 1ug of a mouse 3T3 cell RIPA lysate.
Immunoblot Analysis: 0.5-2ug/ml detects Cbl (120kD) in RIPA lysates from human Jurkat and mouse 3T3 cells. 3T3 cell lysate was resolved by electro-phoresis, transferred to nitrocellulose and probed with anti-Cbl (1ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.
Applications are based on unconjugated antibody.

GST fusion protein corresponding to residues 878-906 of human Cbl. This sequence is identical in mouse.

Preparation and Storage
Store at 4 degree C for short-term. For long-term storage, aliquot to avoid repeated freezing and thawing and freeze at -20 degree C. Aliquots are stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

NCBI Description

This gene is a proto-oncogene that encodes a RING finger E3 ubiquitin ligase. The encoded protein is one of the enzymes required for targeting substrates for degradation by the proteasome. This protein mediates the transfer of ubiquitin from ubiquitin conjugating enzymes (E2) to specific substrates. This protein also contains an N-terminal phosphotyrosine binding domain that allows it to interact with numerous tyrosine-phosphorylated substrates and target them for proteasome degradation. As such it functions as a negative regulator of many signal transduction pathways. This gene has been found to be mutated or translocated in many cancers including acute myeloid leukaemia. Mutations in this gene are also the cause of Noonan syndrome-like disorder. [provided by RefSeq, Mar 2012]

Recognizes activated receptor tyrosine kinases, including PDGFA, EGF and CSF1, and terminates signaling. Participates in signal transduction in hematopoietic cells.
