Related Product Information for anti-CASP10 antibody
There are two types of apoptotic caspases, initiator (apical) caspases and effector (executioner) caspases. Caspase 10 is one of the initiator caspases that cleave inactive pro-forms of effector caspases, thereby activating them. Four isoforms, namely Caspase-10a,-10 b,-10c,and -10d have been identified. They derived from a single chain polypeptide proenzyme but have different mature large and small subunits by cleavage. After the death ligand-receptor binding, caspade-10 has joined the multimeric Fas/TNF receptor complex and gets cleaved. The cleaved caspase-10 further acts on other effector caspases to initiate the caspase cascade.

NCBI Description

This gene encodes a protein which is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. This protein cleaves and activates caspases 3 and 7, and the protein itself is processed by caspase 8. Mutations in this gene are associated with type IIA autoimmune lymphoproliferative syndrome, non-Hodgkin lymphoma and gastric cancer. Alternatively spliced transcript variants encoding different isoforms have been described for this gene. [provided by RefSeq, Apr 2011]

Uniprot Description

Function: Involved in the activation cascade of caspases responsible for apoptosis execution. Recruited to both Fas- and TNFR-1 receptors in a FADD dependent manner. May participate in the granzyme B apoptotic pathways. Cleaves and activates caspase-3, -4, -6, -7, -8, and -9. Hydrolyzes the small- molecule substrates, Tyr-Val-Ala-Asp-|-AMC and Asp-Glu-Val-Asp-|-AMC. Ref.12Isoform C is proteolytically inactive. Ref.12

Catalytic activity: Strict requirement for Asp at position P1 and has a preferred cleavage sequence of Leu-Gln-Thr-Asp-|-Gly.

Subunit structure: Heterotetramer that consists of two anti-parallel arranged heterodimers, each one formed by a 23/17 kDa (p23/17) (depending on the splicing events) and a 12 kDa (p12) subunit

By similarity. Self-associates. Interacts with FADD and CASP8. Found in a Fas signaling complex consisting of FAS, FADD, CASP8 and CASP10. Ref.12

Tissue specificity: Detectable in most tissues. Lowest expression is seen in brain, kidney, prostate, testis and colon.

Post-translational modification: Cleavage by granzyme B and autocatalytic activity generate the two active subunits.

Involvement in disease: Autoimmune lymphoproliferative syndrome 2A (ALPS2A) [MIM:603909]: A disorder of apoptosis that manifests in early childhood and results in the accumulation of autoreactive lymphocytes. It is characterized by non-malignant lymphadenopathy with hepatosplenomegaly, and autoimmune hemolytic anemia, thrombocytopenia and neutropenia.Note: The disease is caused by mutations affecting the gene represented in this entry. Ref.14 Ref.17Familial non-Hodgkin lymphoma (NHL) [MIM:605027]: Cancer that starts in cells of the lymph system, which is part of the body's immune system. NHLs can occur at any age and are often marked by enlarged lymph nodes, fever and weight loss.Note: The gene represented in this entry is involved in disease pathogenesis. Ref.15Gastric cancer (GASC) [MIM:613659]: A malignant disease which starts in the stomach, can spread to the esophagus or the small intestine, and can extend through the stomach wall to nearby lymph nodes and organs. It also can metastasize to other parts of the body. The term gastric cancer or gastric carcinoma refers to adenocarcinoma of the stomach that accounts for most of all gastric malignant tumors. Two main histologic types are recognized, diffuse type and intestinal type carcinomas. Diffuse tumors are poorly differentiated infiltrating lesions, resulting in thickening of the stomach. In contrast, intestinal tumors are usually exophytic, often ulcerating, and associated with intestinal metaplasia of the stomach, most often observed in sporadic disease.Note: The gene represented in this entry is involved in disease pathogenesis. Ref.16

Sequence similarities: Belongs to the peptidase C14A family.Contains 2 DED (death effector) domains.

Product Notes

The CASP10 casp10 (Catalog #AAA648344) is an Antibody produced from Rat and is intended for research purposes only. The product is available for immediate purchase. The CASP10, CT (Caspase-10, CASP-10, ICE-like Apoptotic Protease 4, Apoptotic Protease Mch-4, FAS-associated Death Domain Protein Interleukin-1B-converting Enzyme 2, FLICE2, Caspase-10 Subunit p23/17, Caspase-10 Subunit p12, MCH4) reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's CASP10 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Suitable for use in Western Blot. Dilution: Western Blot: 1-2ug/ml. Researchers should empirically determine the suitability of the CASP10 casp10 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CASP10, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
