ELISA (EL/EIA), Western Blot (WB), Immunoprecipitation (IP)
Application Notes
Suitable for use in ELISA, Western Blot, Immunoprecipitation and Inibition.
Dilution: Direct ELISA: 0.5-1.0ug/ml. The detection limit for recombinant human Brevican is ~1ng/well.
Western Blot: 1-2ug/ml. The detection limit for recombinant human Brevican is ~10ng/lane under non-reducing conditions.
Immunoprecipitation: 25ug/ml immunoprecipitates recombinant human Brevican from conditioned cull culture medium.
Inhibition (Blocking of receptor-ligand interaction): ~0.01-0.05ug/ml will block 50% of the binding of 1ug/ml of Hyaluronan (132kD) to immobilized recombinant human Brevican (100ul of a 3ug/ml solution was coated in each well) in a functional ELISA. 1ug/ml will achieve 100% blocking in the assay.
Immunogen
Recombinant protein corresponding to aa23-911 from human Brevican expressed in NSO cells (Q96GW7).
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Related Product Information for anti-BCAN antibody
Brain-enriched hyaluronan-binding protein (Brevican) is a 160kD member of the aggregan/versican proteoglycan family of matrix proteins. It is brain-derived and serves as a linker between HA and other matrix molecules such as tenascins and fibulins. Mature human Brevican is 889 amino acids in length. It contains multiple domains, with an N-terminal V-type Ig-like domain (aa 52-139) and a C-terminal EGF-like (aa 646-682) and C-type lectin domain (aa 695-802). Additional isoforms exist resulting from an alternate start site at Met228 and a 23 aa substitution for aa 649-911 that serves as a GPI-anchor motif. Over aa 23-911, human Brevican shares 82% aa identity with mouse Brevican.

NCBI Description

This gene encodes a member of the lectican family of chondroitin sulfate proteoglycans that is specifically expressed in the central nervous system. This protein is developmentally regulated and may function in the formation of the brain extracellular matrix. This protein is highly expressed in gliomas and may promote the growth and cell motility of brain tumor cells. Alternate splicing results in multiple transcript variants. [provided by RefSeq, Sep 2011]

Uniprot Description

BCAN: May play a role in the terminally differentiating and the adult nervous system during postnatal development. Could stabilize interactions between hyaluronan (HA) and brain proteoglycans. Belongs to the aggrecan/versican proteoglycan family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Membrane protein, GPI anchor; Secreted, signal peptide; Secreted

Chromosomal Location of Human Ortholog: 1q31

Cellular Component: proteinaceous extracellular matrix; lysosomal lumen; Golgi lumen; extracellular region

Molecular Function: extracellular matrix structural constituent; hyaluronic acid binding; carbohydrate binding

Biological Process: chondroitin sulfate metabolic process; extracellular matrix disassembly; extracellular matrix organization and biogenesis; chondroitin sulfate biosynthetic process; central nervous system development; glycosaminoglycan metabolic process; hippocampus development; chondroitin sulfate catabolic process; carbohydrate metabolic process; pathogenesis; cell adhesion; skeletal development; dermatan sulfate biosynthetic process

Product Notes

The BCAN bcan (Catalog #AAA603829) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Brevican (BEHAB, Brain-Enriched Hyaluronan-Binding Protein) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Brevican can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB), Immunoprecipitation (IP). Researchers should empirically determine the suitability of the BCAN bcan for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Brevican, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
