Members of the Bcl-2 family are pro- or antiapoptotic proteins that process one or several of the Bcl-2 homology (BH) domains. Bid is a proapoptotic, BH3-domain-only member or BH3-only protein (BOP) of the Bcl-2 family. Bid is localized in the cytosolic fraction of living cells as an inactive precursor. Activation of Bid occurs upon its cleavage by caspase-8 in the Fas and TNF- alpha apoptotic signaling pathways. Recent studies have suggested that Bid is also cleaved and activated by granzyme B, calpain and lysosomal extracts. Cleaved Bid translocates to mitochondria where it induces the release of cytochrome c from the intermembrane space of the mitochondria. The release of cytochrome c initiates apoptosis by activating downstream caspases through binding to Apaf-1. Bid has also been shown to interact with with Bax, another Bcl-2 family protein, to trigger a conformational change in Bax leading to the dimerization and integration of Bax into the outer mitochondrial membrane and activating the release of cytochrome c.1-10

Characterization
To insure lot-to-lot consistency, each batch of product is tested by western blotting to conform with the characteristics of a standard reference reagent.
Preparation and Storage
The purified antibody (UNLB) is supplied as 0.1 mg purified immunoglobulin in 0.2 mL of 100 mM borate buffered saline, pH 8.2. No preservatives or amine-containing buffer salts added. Store at 2- 8 degree C.
Reagents are stable for the period shown on the label if stored as directed.

NCBI Description

This gene encodes a death agonist that heterodimerizes with either agonist BAX or antagonist BCL2. The encoded protein is a member of the BCL-2 family of cell death regulators. It is a mediator of mitochondrial damage induced by caspase-8 (CASP8); CASP8 cleaves this encoded protein, and the COOH-terminal part translocates to mitochondria where it triggers cytochrome c release. Multiple alternatively spliced transcript variants have been found, but the full-length nature of some variants has not been defined. [provided by RefSeq]

Post-translational modification: TNF-alpha induces a caspase-mediated cleavage of p22 BID into a major p15 and minor p13 and p11 products
