Specificity
The beta 2-microglobulin subunit (Mr ~ 14 kDa) of avian MHC Class I molecules
Like their mammalian counterparts, avian MHC Class I molecules (also known as B-F antigens) consist of a highly polymorphic alpha -chain (Mr ~ 44 kDa) non-covalently bound to the invariant beta 2-microglobulin subunit (Mr ~ 14 kDa). MHC Class I molecules are expressed on most nucleated cells where they present endogenously synthesized antigenic peptides to CD8+ T lymphocytes, which are usually cytotoxic T cells.1-2 MAb F21-21 also reacts with turkey beta 2-microglobulin.3
Form/Format
Purified (UNLB) Antibody
Sequence Length
119
Applicable Applications for anti-B2M antibody
Flow Cytometry; Immunohistochemistry; Immunoprecipitation; Western Blot; Immunoaffinity Chromatography
Application Notes
Flow Cytometry Suggested Dilution: <= 1 ug/10^6 cells
Characterization
To ensure optimal performance, each batch of monoclonal antibody is tested by flow cytometry to conform to characteristics of a standard reference reagent. Representative flow data are included on page 2 of this product insert.
Preparation and Storage
The purified (UNLB) antibody is supplied as 0.5 mg of purified immunoglobulin in 1.0 mL of 100 mM borate buffered saline, pH 8.2. No preservatives or amine-containing buffer salts added. Store at 2- 8 degree C.
The fluorescein (FITC) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8 degree C.
The biotin (BIOT) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8 degree C.
The R-phycoerythrin (R-PE) conjugate is supplied as 0.1 mg in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C. Do not freeze!
Protect conjugated forms from light. Each reagent is stable for the period shown on the bottle label if stored as directed.

NCBI Description

This gene encodes a serum protein found in association with the major histocompatibility complex (MHC) class I heavy chain on the surface of nearly all nucleated cells. The protein has a predominantly beta-pleated sheet structure that can form amyloid fibrils in some pathological conditions. A mutation in this gene has been shown to result in hypercatabolic hypoproteinemia.

Uniprot Description

Function: Component of the class I major histocompatibility complex (MHC). Involved in the presentation of peptide antigens to the immune system.

Subunit structure: Heterodimer of an alpha chain and a beta chain. Beta-2-microglobulin is the beta-chain of major histocompatibility complex class I molecules. Polymers of beta 2-microglobulin can be found in tissues from patients on long-term hemodialysis. Ref.14

Subcellular location: Secreted. Note: Detected in serum and urine. Ref.16 Ref.17

Post-translational modification: Glycation of Ile-21 is observed in long-term hemodialysis patients.

Involvement in disease: Defects in B2M are the cause of hypercatabolic hypoproteinemia (HYCATHYP) [

MIM:241600]. Affected individuals show marked reduction in serum concentrations of immunoglobulin and albumin, probably due to rapid degradation. Ref.33Note=Beta-2-microglobulin may adopt the fibrillar configuration of amyloid in certain pathologic states. The capacity to assemble into amyloid fibrils is concentration dependent. Persistently high beta(2)-microglobulin serum levels lead to amyloidosis in patients on long-term hemodialysis. Ref.13 Ref.14 Ref.15 Ref.16 Ref.17 Ref.25 Ref.26 Ref.27 Ref.28 Ref.29 Ref.30 Ref.31 Ref.32

Sequence similarities: Belongs to the beta-2-microglobulin family.Contains 1 Ig-like C1-type (immunoglobulin-like) domain.

Product Notes

The B2M b2m (Catalog #AAA673936) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Mouse Anti-Chicken b2-microglobulin-UNLB reacts with Chicken and may cross-react with other species as described in the data sheet. AAA Biotech's beta 2-Microglobulin can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry; Immunohistochemistry; Immunoprecipitation; Western Blot; Immunoaffinity Chromatography. Flow Cytometry Suggested Dilution: <= 1 ug/10^6 cells. Researchers should empirically determine the suitability of the B2M b2m for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "beta 2-Microglobulin, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
