IHC Suggested Protocol
1. Tissues are fixed with 10% neutral buffered formalin, cut to ~4um, and placed on a positively charged glass slide.
2. The prepared slide is baked for at least 30 minutes and at most 24 hours at 55-65 degree C.
3. Pretreatment - Perform heat-induced epitope retrieval (HIER) at pH 6 for 10 to 30 minutes.
4. Block – Block with peroxidase solution for 5 minutes at RT.
5. Primary Ab – Dilute antibodies 1:100 – 1:200 and incubate for 10 - 30 minutes at RT.
6. Secondary Ab - Incubate for 20 - 30 minutes at RT.
7. Substrate - Incubate DAB or Fast Red for 5 - 10 minutes at RT.
8. Counterstain - Counterstain with hematoxylin for 30 seconds to 5 minutes, rinse with distilled water and blueing solution for 30 seconds.
9. Dehydrate and apply coverslip.
(Each researcher should optimize the protocol for their experimental model system.)
Preparation and Storage
Store at 2-8 degree C. Do not freeze.

Annexin A1 is a member of the annexin family, a group of phospholipid-binding membrane proteins that plays a role in both the innate and adaptive immune system. Its plays an important role in controlling inflammation, prostaglandins, and leukotriene mediators. Annexin A1 is overexpressed in 97% of hairy cell leukemia samples, and is absent in other B cell lymphomas.

NCBI Description

This gene encodes a membrane-localized protein that binds phospholipids. This protein inhibits phospholipase A2 and has anti-inflammatory activity. Loss of function or expression of this gene has been detected in multiple tumors. [provided by RefSeq, Dec 2014]

Plays a role in glucocorticoid-mediated down-regulation of the early phase of the inflammatory response (). Promotes resolution of inflammation and wound healing (PubMed:25664854). Functions at least in part by activating the formyl peptide receptors and downstream signaling cascades (PubMed:15187149, PubMed:25664854). Promotes chemotaxis of granulocytes and monocytes via activation of the formyl peptide receptors (PubMed:15187149). Contributes to the adaptive immune response by enhancing signaling cascades that are triggered by T-cell activation, regulates differentiation and proliferation of activated T-cells (PubMed:17008549). Promotes the differentiation of T-cells into Th1 cells and negatively regulates differentiation into Th2 cells (PubMed:17008549). Has no effect on unstimulated T cells (PubMed:17008549). Promotes rearrangement of the actin cytoskeleton, cell polarization and cell migration (PubMed:15187149). Negatively regulates hormone exocytosis via activation of the formyl peptide receptors and reorganization of the actin cytoskeleton (PubMed:19625660). Has high affinity for Ca2+ and can bind up to eight Ca2+ ions (). Displays Ca2+-dependent binding to phospholipid membranes (PubMed:2532504, PubMed:8557678). Plays a role in the formation of phagocytic cups and phagosomes. Plays a role in phagocytosis by mediating the Ca2+-dependent interaction between phagosomes and the actin cytoskeleton ().
