Host
Mouse
Reactivity
Bovine, Chicken, Human, Rabbit
Clonality
Monoclonal
Isotype

Specificity
Recognizes fetal cardiac actin. Represents an excellent marker for cardiac tissue; it discriminates fetal (cardiac) a-actin from all other actin isoforms. Shows no crossreaction with other actin isoforms present in skeletal and smooth muscle, provided that stringent experimental conditions have been applied.
Purity/Purification
Affinity Purified
Purified by Protein G affinity chromatography.
Form/Format
Supplied as a lyophilized powder from PBS, pH 7.4, 0.5% BSA, 0.09% sodium azide. Reconstitute with 1ml sterile ddH2O.
Applicable Applications for anti-ACTN1 antibody
ELISA (EL/EIA), Western Blot (WB), Immunohistochemistry (IHC)
Application Notes
Suitable for use in Western Blot, ELISA and Immunohistochemistry.
Dilution: ELISA: 1:500 (see Note below).
Western Blot: 1:1000 (using the ECL-enhanced procedure)
Note: It is necessary to include 1M sodium chloride for application in Western Blot and ELISA to enhance specificity.
Immunohistochemistry (frozen and paraffin): 1:10. It is necessary to include 0.5M sodium chloride in all washing buffers for application on frozen tissue. This is not necessary for paraffin sections. Microwave treatment is necessary for paraffin sections.
Immunogen
Synthetic N-terminus decapeptide of cardiac isoform of actin.
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

NCBI Description

Alpha actinins belong to the spectrin gene superfamily which represents a diverse group of cytoskeletal proteins, including the alpha and beta spectrins and dystrophins. Alpha actinin is an actin-binding protein with multiple roles in different cell types. In nonmuscle cells, the cytoskeletal isoform is found along microfilament bundles and adherens-type junctions, where it is involved in binding actin to the membrane. In contrast, skeletal, cardiac, and smooth muscle isoforms are localized to the Z-disc and analogous dense bodies, where they help anchor the myofibrillar actin filaments. This gene encodes a nonmuscle, cytoskeletal, alpha actinin isoform and maps to the same site as the structurally similar erythroid beta spectrin gene. Three transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

Product Notes

The ACTN1 actn1 (Catalog #AAA601397) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Actin, Cardiac Fetal reacts with Bovine, Chicken, Human, Rabbit and may cross-react with other species as described in the data sheet. AAA Biotech's Actin, Cardiac Fetal can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB), Immunohistochemistry (IHC). Suitable for use in Western Blot, ELISA and Immunohistochemistry. Dilution: ELISA: 1:500 (see Note below). Western Blot: 1:1000 (using the ECL-enhanced procedure) Note: It is necessary to include 1M sodium chloride for application in Western Blot and ELISA to enhance specificity. Immunohistochemistry (frozen and paraffin): 1:10. It is necessary to include 0.5M sodium chloride in all washing buffers for application on frozen tissue. This is not necessary for paraffin sections. Microwave treatment is necessary for paraffin sections. Researchers should empirically determine the suitability of the ACTN1 actn1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Actin, Cardiac Fetal, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
