Testing Data (Figure 1: Binding of ACE2 (human) to the Spike protein of SARS-CoV-2 is inhibited by the antibody anti-ACE2 (human), mAb (blocking) (AC384) (29-1005). Methods: Spike (SARS-CoV-2):Fc (human) (RBD) (rec.) (32-190018) is coated on an ELISA plate at 1ug/ml. ACE2 (human), mAb (blocking) (AC384) (29-1005) or an unrelated mAb Control are added (starting at 40ug/ml with a two-fold serial dilution) together with 500ng/ul of ACE2 (human) (32-190010). After incubation for 1h at RT, the binding was detected using an anti-DYKDDDDK antibody (HRP).)

Related Product Information for anti-ACE-2 antibody
Angiotensin-converting enzyme2 (ACE2) is an ectoenzyme (carboxypeptidase) with an extracellular catalytic domain that predominantly localizes at the plasma membrane and is thereby able to hydrolyze circulating peptides. ACE2 has approximately 42% sequence identity with ACE and its cytoplasmic and transmembrane domains show 48% homology to the protein collectrin, that plays a critical role in the amino acid absorption of the kidney. ACE2 converts angiotensin I to angiotensin 1-9, a peptide of unknown function, and angiotensin II to angiotensin 1-7, a vasodilator. ACE2 is involved in the regulation of systemic blood pressure and has direct effects on cardiac functions. It is expressed predominantly in endothelial cells of the lung, gut, heart and kidney. ACE2 together with the protease TMPRSS2 acts as a functional receptor for SARS coronavirus as well as for the new highly pathogenic coronavirus, 2019-nCoV/SARS-CoV-2, which is cause for pneumonia COVID-19.

NCBI Description

The protein encoded by this gene belongs to the angiotensin-converting enzyme family of dipeptidyl carboxydipeptidases and has considerable homology to human angiotensin 1 converting enzyme. This secreted protein catalyzes the cleavage of angiotensin I into angiotensin 1-9, and angiotensin II into the vasodilator angiotensin 1-7. The organ- and cell-specific expression of this gene suggests that it may play a role in the regulation of cardiovascular and renal function, as well as fertility. In addition, the encoded protein is a functional receptor for the spike glycoprotein of the human coronavirus HCoV-NL63 and the human severe acute respiratory syndrome coronaviruses, SARS-CoV and SARS-CoV-2 (COVID-19 virus). Uniprot Description

ACE2: Carboxypeptidase which converts angiotensin I to angiotensin 1-9, a peptide of unknown function, and angiotensin II to angiotensin 1-7, a vasodilator. Also able to hydrolyze apelin- 13 and dynorphin-13 with high efficiency. May be an important regulator of heart function. In case of human coronaviruses SARS and HCoV-NL63 infections, serve as functional receptor for the spike glycoprotein of both coronaviruses. Interacts with ITGB1. Interacts with SARS-CoV and HCoV- NL63 spike glycoprotein. Up-regulated in failing heart. Expressed in endothelial cells from small and large arteries, and in arterial smooth muscle cells. Expressed in lung alveolar epithelial cells, enterocytes of the small intestine, Leydig cells and Sertoli cells. Expressed in heart, kidney, testis, and gastrointestinal system. Activated by chloride and fluoride, but not bromide. Inhibited by MLN-4760, cFP_Leu, and EDTA, but not by the ACE inhibitors linosipril, captopril and enalaprilat. Belongs to the peptidase M2 family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The ACE-2 ace2 (Catalog #AAA669409) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The anti-ACE2 (human), mAb (blocking) (AC384) Azide free reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's ACE-2 can be used in a range of immunoassay formats including, but not limited to, Functional Assay, ELISA (EIA), Western Blot (WB). ELISA: direct or indirect: 1:2,000-1:10,000 WB: 1:2,000-1:10,000 using ECL. Suggested blocking and dilution buffer is PBST with 0.05% Tween 20 and 5% skim milk. Functional Application: Blocks the binding of human ACE2 to the Spike protein of SARS-Cov-2. Researchers should empirically determine the suitability of the ACE-2 ace2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ACE-2, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
