Inhibition of Laminin Binding to Dystroglycan:In anitrocellulose overlay experiment, this antibody inhibits bindingof 125I Laminin to Dystroglycan.Western Blot: 1:1000-1:2000 Detects alpha-Dystroglycan inrabbit skeletal muscle. Rabbit skeletal muscle lysate wasresolved by electrophoresis, transferred to nitrocellulose andprobed with D9905-05. Proteins were visualized using an IgG(HRP) goat anti-mouse and a chemiluminescence detectionsystem.Note:The use of WGA purified protein results in significantlycleaner blots and immunoprecipitates. Post-translationalmodification of Dystroglycan causes band broadening. Optimal dilutions to be determined by the researcher. Other applications not tested.

Rabbit skeletal muscle membrane preparation.
Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Dystroglycans are essential elements of the neuromuscularjunction (NMJ). The gene for dystroglycan is expressed as aprecursor protein that is post translationally cleaved into a156kD extracellular peripheral membrane protein called alphadystroglycan and a 43kD transmembrane protein, betadystroglycan. The latter protein contains a PPxY motif thatpromotes binding to WW domain containing proteins, such asutrophin and dystrophin. Phosphorylation at tyrosine 892within the PPxY motif may regulate c-Src interactions with betadystroglycan, as well as inhibit interactions with WW domainproteins. In skeletal muscle, beta dystroglycan is normallylocalized to the plasma membrane, however phosphorylationof Tyr892 leads to localization of beta dystroglycan toendosomal compartments along with c-Src. Thus,phosphorylation at Tyr892 may have important roles in alteringthe localization of beta dystroglycan during NMJ formation

Researchers should empirically determine the suitability of the a-Dystroglycan for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "a-Dystroglycan, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
