Dilution: Immunoprecipitation: 2ug/mg protein lysate. Use Protein G.
Western Blot: 1ug/ml for 2 hours at RT.
Immunohistochemistry (Formalin/paraffin): 2-4ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 minutes followed by cooling at RT for 20 minutes.
Western Blot (WB)
(Western Blot analysis of whole cell extracts from different cell lines using antibody MBS632037. Samples were electrophoresced usng Novex NuPAGE 10% Bris-Tris gel. Resolved proteins were transferred onto a nitrocellulose membrane and blocked with 5% skim milk. Detection was by chemiluminescence using IgG (HRP) goat anti-mouse secondary antibody and Novex ECL chemiluminescent reagent kit (Pierce Cat. # WP20005).)

(Immunohistochemistry analysis of formalin-fixed, paraffin-embedded human skin using antibody MBS632037. Peroxidase-conjugated secondary antibody and AEC staining were used for visualization. Note cytoplasmic staining of epidermis.)

Binds to a large number of partners, usually by recognition of a phosphoserine or phosphothreonine motif. Binding generally results in the modulation of the activity of the binding partner. When bound to KRT17, regulates protein synthesis and epithelial cell growth by stimulating Akt/mTOR pathway. Homodimer. Interacts with KRT17 and SAMSN1. Found in a complex with XPO7, EIF4A1, ARHGAP1, VPS26A, VPS29, VPS35 and SFN. Interacts with GAB2. Interacts with SRPK2. Present mainly in tissues enriched in stratified squamous keratinizing epithelium. Belongs to the 14-3-3 family. 2 isoforms of the human protein are produced by alternative splicing.
