NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between IL-4Ralpha and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This IL-4Ralpha ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey IL-4Ralpha. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: IL-4Ralpha ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-IL-4Ralpha antibody and an IL-4Ralpha-HRP conjugate. The assay sample and buffer are incubated together with IL-4Ralpha-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-4Ralpha concentration since IL-4Ralpha from samples and IL-4Ralpha-HRP conjugate compete for the anti-IL-4Ralpha antibody binding site. Since the number of sites is limited, as more sites are occupied by IL-4Ralpha from the sample, fewer sites are left to bind IL-4Ralpha-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-4Ralpha concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes the alpha chain of the interleukin-4 receptor, a type I transmembrane protein that can bind interleukin 4 and interleukin 13 to regulate IgE production. The encoded protein also can bind interleukin 4 to promote differentiation of Th2 cells. A soluble form of the encoded protein can be produced by proteolysis of the membrane-bound protein, and this soluble form can inhibit IL4-mediated cell proliferation and IL5 upregulation by T-cells. Allelic variations in this gene have been associated with atopy, a condition that can manifest itself as allergic rhinitis, sinusitus, asthma, or eczema. Polymorphisms in this gene are also associated with resistance to human immunodeficiency virus type-1 infection. Alternate splicing results in multiple transcript variants.
