Monkey Butyrophilin-like protein 2 (BTNL2) ELISA Kit
Principle of the Assay||BTNL2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-BTNL2 antibody and an BTNL2-HRP conjugate. The assay sample and buffer are incubated together with BTNL2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BTNL2 concentration since BTNL2 from samples and BTNL2-HRP conjugate compete for the anti-BTNL2 antibody binding site. Since the number of sites is limited, as more sites are occupied by BTNL2 from the sample, fewer sites are left to bind BTNL2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BTNL2 concentration in each sample is interpolated from this standard curve.

BTNL2: Negative regulator of T-cell proliferation. Genetic variation in BTNL2 is a cause of susceptibility to sarcoidosis type 2 (SS2). Sarcoidosis is an idiopathic, systemic, inflammatory disease characterized by the formation of immune granulomas in involved organs. Granulomas predominantly invade the lungs and the lymphatic system, but also skin, liver, spleen, eyes and other organs may be involved. A nucleotide transition affecting a splice donor site results in the use of an alternative splice site and the production of isoform 3. Individuals expressing isoform 3 have a higher risk for sarcoidosis. Belongs to the immunoglobulin superfamily. BTN/MOG family. 6 isoforms of the human protein are produced by alternative splicing.
