Principle of the Assay||This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MMP-14 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MMP-14 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MMP-14 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MMP-14 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Function: Seems to specifically activate progelatinase A. May thus trigger invasion by tumor cells by activating progelatinase A on the tumor cell surface May be involved in actin cytoskeleton reorganization by cleaving PTK7 Acts as a positive regulator of cell growth and migration via activation of MMP15. Involved in the formation of the fibrovascular tissues

Catalytic activity: Endopeptidase activity. Activates progelatinase A by cleavage of the propeptide at 37-Asn-|-Leu-38. Other bonds hydrolyzed include 35-Gly-|-Ile-36 in the propeptide of collagenase 3, and 341-Asn-|-Phe-342, 441-Asp-|-Leu-442 and 354-Gln-|-Thr-355 in the aggrecan interglobular domain.

Cofactor: Binds 1 zinc ion per subunit
