Intended Uses: For the quantitative detection of Horse Collagenase 3 (MMP13) concentration in serum, plasma, tissue homogenates and other biological fluids.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate MMP13 in Horse serum, plasma, tissue homogenates. An antibody specific for MMP13 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MMP13 present is bound by the immobilized antibody. After removing any unbound substances, a biotin- conjugated antibody specific for MMP13 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MMP13 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Uniprot Description

Plays a role in the degradation of extracellular matrix proteins including fibrillar collagen, fibronectin, TNC and ACAN. Cleaves triple helical collagens, including type I, type II and type III collagen, but has the highest activity with soluble type II collagen. Can also degrade collagen type IV, type XIV and type X. May also function by activating or degrading key regulatory proteins, such as TGFB1 and CTGF. Plays a role in wound healing, tissue remodeling, cartilage degradation, bone development, bone mineralization and ossification. Required for normal embryonic bone development and ossification. Plays a role in the healing of bone fractures via endochondral ossification. Plays a role in wound healing, probably by a mechanism that involves proteolytic activation of TGFB1 and degradation of CTGF. Plays a role in keratinocyte migration during wound healing. May play a role in cell migration and in tumor cell invasion ().
