Principle of the Assay||This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MME has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MME present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MME is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MME bound in the initial step. The color development is stopped and the intensity of the color is measured.

Function: Thermolysin-like specificity, but is almost confined on acting on polypeptides of up to 30 amino acids. Biologically important in the destruction of opioid peptides such as Met- and Leu-enkephalins by cleavage of a Gly-Phe bond. Able to cleave angiotensin-1, angiotensin-2 and angiotensin 1-9 Involved in the degradation of atrial natriuretic factor (ANF). Displays UV-inducible elastase activity toward skin preelastic and elastic fibers

Catalytic activity: Preferential cleavage of polypeptides between hydrophobic residues, particularly with Phe or Tyr at P1'.
