Principle of the Assay: MK ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-MK antibody and an MK-HRP conjugate. The assay sample and buffer are incubated together with MK-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MK concentration since MK from samples and MK-HRP conjugate compete for the anti-MK antibody binding site. Since the number of sites is limited, as more sites are occupied by MK from the sample, fewer sites are left to bind MK-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MK concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a member of a small family of secreted growth factors that binds heparin and responds to retinoic acid. The encoded protein promotes cell growth, migration, and angiogenesis, in particular during tumorigenesis. This gene has been targeted as a therapeutic for a variety of different disorders. Alternatively spliced transcript variants encoding multiple isoforms have been observed. [provided by RefSeq, Jul 2012]
Uniprot Description
MDK: Developmentally regulated, secreted growth factor homologous to pleiotrophin (PTN), which has heparin binding activity. Binds anaplastic lymphoma kinase (ALK) which induces ALK activation and subsequent phosphorylation of the insulin receptor substrate (IRS1), followed by the activation of mitogen-activated protein kinase (MAPK) and PI3-kinase, and the induction of cell proliferation. Involved in neointima formation after arterial injury, possibly by mediating leukocyte recruitment. Also involved in early fetal adrenal gland development. Belongs to the pleiotrophin family.

Product Notes
The Monkey MK mdk (Catalog #AAA743873) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA743873 ELISA Kit recognizes Monkey MK. It is sometimes possible for the material contained within the vial of "Midkine, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
