Principle of the Assay: MCC ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-MCC antibody and an MCC-HRP conjugate. The assay sample and buffer are incubated together with MCC-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MCC concentration since MCC from samples and MCC-HRP conjugate compete for the anti-MCC antibody binding site. Since the number of sites is limited, as more sites are occupied by MCC from the sample, fewer sites are left to bind MCC-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MCC concentration in each sample is interpolated from this standard curve.

This gene product is a chymotryptic serine proteinase that belongs to the peptidase family S1. It is expressed in mast cells and thought to function in the degradation of the extracellular matrix, the regulation of submucosal gland secretion, and the generation of vasoactive peptides. In the heart and blood vessels, this protein, rather than angiotensin converting enzyme, is largely responsible for converting angiotensin I to the vasoactive peptide angiotensin II. Angiotensin II has been implicated in blood pressure control and in the pathogenesis of hypertension, cardiac hypertrophy, and heart failure.
